Abstract Durian Kura-kura ( Durio testudinarius Becc.) belongs to the Malvaceae family and is an endemic species of Borneo. Recently, genomic-based next-generation sequencing (NGS) approaches have been carried out for germplasm conservation plant breeding programs. The NGS technologies allow genomes be sequenced quickly inexpensively enable efficient development SSR markers through in-silico approaches. This study aimed develop characterize simple sequence repeats (SSRs) from assembled genome. 1203929 scaffolds genome were produced Ray assembler. SSRs identified extracted using MISA program 4315 sequences containing SSRs. six motif identified; consist 431 dinucleotide (the most AT), 3257 trinucleotide TTA), 516 tetranucleotide AAAT), 89 pentanucleotide ATTTT), 18 hexanucleotide four heptanucleotide. new will used in further studies genetic population D.
